FDA Grants Fast Track Designation to Investigational Treatment for Cognitive Impairment in Adults with Neuropsychiatric Systemic Lupus Erythematosus
The U.S. Food and Drug Administration (FDA) has granted Fast Track designation to EG-501 from Evergreen Therapeutics, an investigational oral treatment for adults with cognitive impairment associated with neuropsychiatric systemic lupus erythematosus (NPSLE), following promising Phase 2 results from the ClearMEMory study. Currently, there are no approved therapies specifically targeting cognitive impairment in NPSLE.
The 12-week, randomized, double-blind, placebo-controlled Phase 2 trial evaluated EG-501 in 56 adults with lupus-related cognitive dysfunction across three clinical sites, including Vanderbilt Health and Cleveland Clinic. Researchers assessed cognitive performance using standardized neuropsychological testing and found greater improvements among participants receiving EG-501 than among those receiving placebo. The investigational treatment works by targeting NMDA receptors, with preliminary research suggesting potential neuroprotective and anti-inflammatory effects. Detailed effect sizes and P values were not reported.
The designation marks an encouraging regulatory step for EG-501. Evergreen Therapeutics is preparing a larger Phase 3 trial to further evaluate the treatment and confirm its potential benefits, though a timeline has not been announced. While additional research is needed to confirm the findings, the FDA's Fast Track designation could help accelerate development of a targeted treatment for lupus-related cognitive impairment, an area of significant unmet need.
